Super-enhanceri sintetici permit imunoterapia virală de precizie

Conform nature.com, cercetătorii au dezvoltat super-enhanceri sintetici (SSE) pentru imunoterapia virală, având rezultate promițătoare în tratamentul glioblastomului. Acești SSE sunt proiectați să stimuleze selectiv expresia genelor terapeutice în celulele stem glioblastomice, optimizând astfel eficacitatea tratamentului și reducând efectele secundare în țesutul cerebral sănătos.

Promotorii specifici pentru tipul celular sunt utilizați în terapia genică pentru a restricționa expresia încărcăturii terapeutice. Folosind perspective recente asupra funcției enhancerilor, am dezvoltat SSE-uri prin asamblarea fragmentelor validate funcțional în aranjamente multilayer. Concentrați pe rețeaua de reglementare transcripțională SOX2-și SOX9-driven în celulele stem glioblastomice (GSCs), am creat SSE-uri cu activitate robustă și selectivitate ridicată. Profilarea la nivel celular unic, analizele biochimice și datele de legare genomică au arătat că SSE-urile integrează factori de transcripție neurodezvoltare și de semnalizare pentru a declanșa formarea de complexe multimere mari de factori de transcripție.

În plus, expresia selectivă în GSC-uri a unei combinații de încărcături citotoxice (HSV-TK și ganciclovir) și imunomodulatoare (IL-12), livrate prin intermediul vectorilor virusului asociat adenou, a condus la rezultate curative într-un model animal de glioblastom agresiv. Notabil, IL-12 a indus o memorie imunologică care a prevenit recidiva tumorii. Activitatea și selectivitatea virusului asociat adenou și a SSE-urilor au fost validate folosind țesutul primar de glioblastom uman și probe de cortex normal.

În concluzie, SSE-urile valorifică programele de transcripție esențiale care definesc fenotipul GSC și permit activarea precisă a imunității. Această abordare ar putea avea aplicații mai largi în alte contexte, atunci când este necesar controlul precis al expresiei transgene în state celulare specifice.
